不同剂量四君子汤对急性肝衰竭小鼠肠屏障功能的影响

摘    要:目的:探讨四君子汤对急性肝衰竭小鼠肠屏障功能的影响及治疗最佳剂量。方法:小鼠经腹腔一次性注射D-GaIN/LPS,制备急性肝衰竭模型,观察3种剂量(10g/kg、15g/kg、25g/kg)四君子汤对肠道正常菌群,肠黏膜sIgA含量及肝功能的影响。结果:小鼠腹腔注射D-GaIN/LPS后,血清中ALT、AST活性显著提高(P<0.01);肠道内肠杆菌、肠球菌数量明显上升(P<0.01),乳酸杆菌、双歧杆菌数量明显下降(P<0.01);肠黏膜sIgA含量明显降低(P<0.01)。3种剂量组上述指标均得到明显改善,与自然恢复组比较差异有统计学意义(P<0.05,P<0.01);治疗组间比较发现中、高剂量组效果相当,但好于低剂量组。结论:急性肝功能衰竭时肠道菌群失调,肠粘膜免疫功能下降,四君子汤具有一定的调整作用,且中剂量的四君子汤(15g/kg)可能为适宜剂量。

EFFECT OF SIJUNZI DECODTION ON THE INTESTINAL BARRIER FUNCTION OF THE ACUTE LIVER FAILURE MICE

Abstract:Objective:Effect of Sijunzi Decoction on intestinal barrier function was studied in acute liver failure mice.Methods:Acute liver failure was induced in mice with D-galactosamine(D-GalN) and endotoxin(LPS).Acute liver failure mice were treated with Sijunzi Decoction,and the lever of secretary IgA(sIgA) of intestinal mucosa,the changes of intestinal microflora and liver function were determined.Results:After intraperitoneal injection of D-GalN,the activities of ALT and AST in the serum of the model mice increase differently(P<0.01),the numbers of intestinal bacilli and enterococci increase significantly(P<0.01),lactic acid bacteria and,bifidobacteria decrease(P<0.01) the level of sIgA in intestinal mucosa decrease(P<0.01).Compared with natural recovery mice,all the index of the mice treated with Sijunzi Decoction improved(P<0.05,P<0.01) and those of 25 g/kg group are equal with 15 g/kg group.Conclusion:Intestinal barrier is damaged obviously in the acute liver failure mice and Sijunzi Decoction could improve intestinal function.
