亲和色谱分离纯化低分子肝素

摘    要:目的探索亲和色谱纯化低分子肝素的方法。方法以壳聚糖为载体,以抗凝血酶Ⅲ及凝血酶为配基分别制备亲和吸附剂,采用亲和方法分离纯化低分子肝素,并对两者的纯化效果进行比较。结果前者制备的低分子肝素抗Ⅹa因子活性为173.9 IU/mg,比纯化前提高1.7倍;后者制备的低分子肝素抗Ⅹa因子活性为646.0 IU/mg,比纯化前提高6.3倍。结论经壳聚糖-抗凝血酶Ⅲ纯化的低分子肝素其抗Ⅹa因子活性显著提高。

Abstract:Purpose To study purification method of the low molecular weight heparin(LMWH) by affinity chromatography.MethodsChitosan-thrombin and chitosan-antithrombinⅢ adsorbent were prepared respectively.The effects of above two affinity adsorbents were compared through the anti-FⅩa activity of purified LMWH.ResultsIt showed that the anti-FⅩa activities of the LMWH purified through chitosan-thrombin adsorbent and chitosan-antithrombinⅢ adsorbent were 173.9 U/mg and 646.0 U/mg,which raised 1.7 times and 6.3 times respectively compared with the unpurified LMWH.ConclusionThe anti-FⅩa activity of LMWH purified through chitosan-antithrombinⅢ adsorbent was evidently elevated.
