瘦素受体在实验性多囊卵巢综合征大鼠卵巢中的表达及其意义

摘    要:目的研究来曲唑诱导建立多囊卵巢综合征(PCOS)动物模型中卵巢组织瘦素受体(Ob-R)及其mRNA的表达,探讨卵巢Ob-R变化与PCOS发病机制的关系。方法6周龄清洁级雌性SD大鼠45只,分为正常组、溶剂组和来曲唑组,各15只。正常组每日自由摄取食物和水;溶剂组每日给予l%羟甲基纤维素水溶液2mL/kg灌胃;来曲唑组以来曲唑0.5mg/kg灌胃。观察大鼠体质量、动情周期、卵巢质量及其形态学改变,测定血清黄体生成素(LH)、睾酮(T)、胰岛素(INS)、瘦素(LEP)及血糖(FG)水平,采用免疫组织化学和RealTimeRT-PCR方法检测大鼠卵巢中Ob-R及其mRNA的相对表达,并对3组的各指标进行比较。结果来曲唑组大鼠体质量和卵巢质量指数升高,动情周期消失,卵巢体积增大,白膜增厚,H—E染色可见较多囊状扩张的卵泡且黄体数量明显下降,血清T、LH、INS、LEP水平明显升高(P〈0.05);卵巢Ob-R及其mRNA表达明显增强(P〈0.05)。结论来曲唑诱导建立的PCOS动物模型临床表现与人类似;卵巢中Ob—R表达上调,可能是导致PCOS患者不排卵、生育力下降的原因之一。
