桑当降低高尿酸血症小鼠血清尿酸的物质基础初步研究

摘    要:目的初步研究桑当降低高尿酸血症模型小鼠血清尿酸水平的物质基础。方法选择尿酸酶抑制剂氧嗪酸钾盐,对小鼠腹腔注射后造成高尿酸血症模型。用系统溶剂法(极性由小到大,石油醚→氯仿→正丁醇→乙醇→水)提取桑当,制成浸膏,用0.5%羧甲基纤维素钠(CMC-Na)配成混悬液,连续7d给小鼠灌胃。采用尿酸酶-过氧化物酶偶联法测定小鼠血清尿酸水平。结果与空白对照组比较,模型对照组血尿酸水平明显升高(P〈0.05);与模型对照组比较,阳性对照组(别嘌呤醇)、桑当石油醚组、桑当氯仿组、桑当正丁醇组、桑当乙醇组、桑当水提组高尿酸血症小鼠的血尿酸水平均明显降低(P〈0.05)。结论 5种溶剂的桑当提取液均能明显降低高尿酸血症模型小鼠的血尿酸水平,其中正丁醇组较其他组更显著。

Abstract:Objective To preliminarily study the substance of Sangdang on the treatment of mice with hyperuricaemia. Methods Uricase inhibitor potassium oxonate was selected and intraperitoneally injected for constructing the mice model of hyperuricaemia. Sangdang was systematically extracted by solvents (petroleum ether,chloroform,n- butanol,ethyl alcohol,water according to solvents' polarity from small to big). Then the extract was prepared and the suspension liquid was confected by 0. 5% CMC-Na to give intragastric administration on mice for 7 d. URO-PAP method was used to examine the blood uric acid level. Results The blood uric acid level of the model control group was remarkably higher than that of the blank control group( P〈0. 05). Comparing with the model control group,the blood uric acid levels of the positive control (allopurinol),Sangdang petroleum ether,Sangdang chloroform,Sangdang n-butanol,Sangdang ethyl alcohol and Sangdang water groups were all significantly decreased( P〈0. 05) . Conclusion The Sangdang extractions with the five solvents can remarkably decrease the blood uric acid level of the hyperuricaemia model mice,among them the extraction with n-butanol is more significant than the others.
