姜黄素对人乳腺癌MCF-7细胞增殖抑制作用及机制研究

目的:探讨姜黄素对人乳腺癌MCF-7细胞增殖抑制、诱导凋亡作用及其分子作用机制。方法:MTT法检测姜黄素对MCF-7细胞的增殖抑制作用;流式细胞术PI单染法检测细胞周期;Annexin V/PI双染法检测细胞凋亡;RT-PCR法检测凋亡相关基因Bcl-2和Bax的mRNA表达水平。结果:姜黄素对MCF-7细胞生长有明显抑制作用,并呈剂量、时间依赖性;FCM结果显示姜黄素能使MCF-7细胞阻滞在G1/S期;Annexin V/PI双染法验证姜黄素可以诱导细胞凋亡;RT-PCR结果显示Bax mRNA水平明显上调,而Bcl-2的mRNA表达水平降低。结论:姜黄素对人乳腺癌MCF-7细胞增殖具有显著的抑制作用并可诱导细胞凋亡,其作用机制可能与其上调Bax基因表达水平的同时,下调Bcl-2基因表达水平,从而诱导细胞凋亡有关。

Effect of anti-proliferation of curcumin on human breast cancer MCF-7 cells and its mechanism

Objective:To investigate the inhibiting effect,apoptosis and its mechanism of curcumin on human breast cancer MCF-7 cells.Methods:Growth inhibition rates of MCF-7 cells were measured with MTT method.The cell cycle distribution was detected by flow cytometic analysis and apoptosis was detected by PI fluorescent staining.In addition,the expression levels of bcl-2,bax mRNA were analyzed by RT-PCR.Results:Curcumin inhibited the proliferation of MCF-7 cells in a does-and time dependent manner.The cell growth was arrested in G 1/S phase.Fluorescent staining can detect the apoptotic cells.The expression of Bax mRNA was increased and the Bcl-2 was decreased.Conclusion:Curcumin not only inhibits proliferation but also induces apoptosis of MCF-7 cells.The possible mechanism is related with curcumin down regulating bcl-2 but up regulating bax.
