摘    要:〔目的〕建立快速、敏感、特异、高通量、同时检测多种病原体的技术方法。〔方法〕建立基于双抗体夹心免疫学检测的编码微球悬浮芯片和生物素-亲和素ELISA方法,比较2种方法对炭疽芽孢杆菌定量检测以及直接从"白色粉末"样品中检测的能力。〔结果〕悬浮芯片对炭疽杆菌芽孢检测具有很好的敏感性和特异性,最低检出限为4.2×103cfu/ml,动态范围为103-107cfu/ml;生物素-亲和素ELISA最低可以检测到1.4×104 cfu/ml,线性范围为104-107cfu/ml。〔结论〕悬浮芯片方法检测炭疽杆菌芽孢比ELISA方法具有更高的灵敏度和更宽的动态检测范围,在病原菌的早期识别、快速诊断、应对生物恐怖威胁、控制传染病爆发中具有广阔的应用前景。

Abstract:Objective To establish a platform for rapid,sensitive,specific,high-throughput,and simultaneous detection of multiple biothreat-associated agents.Method A suspension array-based sandwich immunoassay and a biotin-streptavidin enzyme-linked immunosorbent assay were developed for exploring the feasibility of quantitative and directly detection of B.anthracis spores in powder samples.Result The newly developed suspension array appeared to be specific and sensitive,with the detection sensitivities of 4.2 × 103 efu/ml for B. anthracis spores, and the dynamic ranges of 103-107 cfu/ml, which were higher than those of corresponding conventional ELISA tests with sensitivities of 1.4 × 104 cfu/ml and the dynamic ranges of 104-107 cfu/ml. Conclusion The suspension array would be useful for early identification,rapid diagnosis,and response for the attack of bioterrorism and outbreak of infectious disease.
