摘    要:目的:了解p16基因在急性髓系白血病(AML)发病中的意义。方法:用聚合酶链反应-单链DNA构象多态性、Northernblot方法分析16例AML细胞p16基因结构及转录水平表达。结果:16例均无p16基因的缺失、突变。初治和复发的12例AML的p16mRNA表达水平明显低于4例长期缓解者及正常对照。1例p53基因突变AML的p16mRNA有明显升高。结论:p16基因在AML发病中可能不占重要地位,而在AML病态造血的维持中起一定作用,在p53突变时其表达反馈性升高,显示一种反馈环路失衡,提示在研究单个基因的基础上观察细胞周期检测点功能具有重要意义。

Abstract:OBJECTIVE: To explore the role of p16 gene in the pathogenesis of AML. METHODS: PCR-SSCP and Northern blot were used to analyse the structure and mRNA expression of p16 gene in 16 cases of AML. RESULTS: No deletion or mutation of p16 gene were found in all of the 16 cases. The p16 mRNA expression in 12 AML at diagnosis or relapse was significantly lower than that in 4 long term remission cases and normal controls. p16 mRNA expression was significantly higher in a case of AML with p53 mutation. CONCLUSION: p16 gene may not play an important role in the pathogenesis of AML. The high p16 expression in the patient with p53 mutation might reflect a kind of inbalance of the feedback cycle.
