摘    要:目的评价三七皂苷Rg1对动物学习记忆功能的影响及其初步作用机理.方法采用东莨菪碱、乙醇、亚硝酸钠复制小鼠和大鼠记忆障碍模型,运用跳台法研究三七皂苷Rg1对动物学习记忆功能的影响,同时探讨其对脑组织乙酰胆碱酯酶(AChE)活性、丙二醛(MDA)及脂褐质水平的影响.结果与东莨菪碱所致记忆获得障碍、亚硝酸钠所致记忆巩固障碍及40%乙醇所致记忆再现障碍的模型组比较,中、高剂量(小鼠200和400mg/kg,大鼠100和200mg/kg)的三七皂苷Rg1均明显减少5min内的错误次数,并显著延长潜伏期.大鼠大脑皮层生化代谢产物含量测定结果表明,与东莨菪碱所致记忆获得障碍模型组相比,中、高剂量(100和200mg/kg)的三七皂苷Rg1均显著降低AChE活性、MDA及脂褐质的含量.结论三七皂苷Rg1能明显改善记忆的获得、促进记忆的巩固、有利于记忆的再现,具有明显的促智作用,其机制可能与降低脑组织AChE活性及改善单胺类神经系统功能、降低脑组织中脂质过氧化物及脂褐质等的含量有关.

Abstract:Objective To study the effects of Notogisenoside-Rg1 on the abilities of learning and memory and its preliminary mechanisms.Methods ICR mice and SD rats were respectively induced by scopolamine hydrobromide,alcohol,and NaNO2 to make dysmnesia models.The tests of springing to platform after electric stimulation was performed to evaluate the effects of Notogisenoside-Rg1 on the abilities of learning and memory. The influence of Notogisenoside-Rg1 was investigated on acetylcholine esterase (AChE),malondiadehyde (MDA) and lipofuscin in brain tissue.Results Notogisenoside-Rg1 (200 and 400 mg/kg for mice,100 and 200 mg/kg for rats) significantly reduced the error times within 5 min in dysmnesia groups (P 0.01,P 0.05) and prolongesd the incubation periods of memory.Notogisenoside-Rg1 decreased the levels of MDA,suppressed the activity of AChE and obviously inhibited the increase of the lipofuscin in brain tissues of rats.Conclusion Notogisenoside-Rg1 showes significantly improving effects on the abilities of learning and memory. The preliminary mechanisms maybe:it can decrease the activity of AChE to deplete the degradation of acetylcholine (Ach) and decrease the level offree radicals in brain tissue resultingin depletingthe influence ofthe accumulation of lipofuscin in brain tissue on the metabolism and function of brain.
