壬基酚对大鼠睾丸抑制素和雄激素结合蛋白表达的影响

摘    要:目的 观察壬基酚对大鼠睾丸抑制素和雄激素结合蛋白的影响。方法 将36只19d龄SD雄性大鼠分为4组,分别腹腔1次注射0、11、44和176mg/kg壬基酚后,于6h、24h和48h每组各处死3只动物,取睾丸提取RNA后,用RT-PCR法对抑制素和雄激素结合蛋白的表达量进行定量分析。结果 雄激素结合蛋白基因的表达量随所给NP剂量的增高而降低,呈剂量效应关系。与对照组比较,在6h各组中,ABP的表达量降低均具有高度显著性(P<0.01);在24h各组中,与对照组比较,44和176mg/kg组的ABP表达降低有显著性(P<0.05或<0.01)。在48h各组中,仅176mg/kg组的降低具有显著性(P<0.05)。24和48h抑制素表达量,在44和176mg/kg剂量组降低,与对照组比较有统计学意义(P<0.01)。结论 壬基酚可以抑制大鼠睾丸Sertoli细胞雄激素结合蛋白的表达。低剂量的壬基酚可能并不影响Sertoli细胞抑制素的表达,但高剂量时,可以使抑制素表达降低。

Abstract:Objective To observe the effects of nonylphenol on inhibin and androgen binding protein(ABP) expressions in the testis of rats.Method 36 rats (19 day aged) were divided randomly into 4 groups. 0, 11,44 and 176 mg/kg of nonylphenol(NP) were given by intraperitoneal injection in each group respectively. And then 3 rats were sacrificed after 6, 24, and 48 hours in each groups respectively. Abstracted the total RNA from testes and quantified the mRNA of inhibin and ABP in each group by RT-PCR.Results The ABP expressions decreased with the increasing given NP, and there was a dose-effect relationship. Compared with control group, the decreases of ABP in 11,44,176 mg/kg groups were significant (P<0.01) after 6 hours,. in 44 and 176 mg/kg groups were significant after 24hours (P<0.05 or P<0.01), and only in 176 group were significant after 48 hours. The expressions of inhibin were decreased in 44 and 176mg/kg groups respectively (P<0.01) after 24 and 48 hours.Conclusions NP could decrease the expressions of ABP in Sertoli cells of rats. At lower dose, NP might not affect the expressions of inhibin in Sertoli cells, but at a higher dose, it might decrease its expression.
