半夏泻心汤对食管炎大鼠食管平滑肌收缩调控蛋白基因和细胞内游离钙的影响

摘    要:目的:从食管运动角度探讨半夏泻心汤防治反流性食管炎的机制。方法:60只大鼠随机分为伪手术组、模型组、阳性对照组(西沙必利)和半夏泻心汤组(BX)。采用胃-十二指肠混合反流模型;应用多导生理信号记录仪观察离体食管肌条收缩情况;采用RT-PCR的方法观察食管平滑肌收缩调控蛋白基因表达情况;激光共聚焦的方法观察细胞内游离钙的变化。结果:与模型组相比,半夏泻心汤组大鼠食管离体肌条收缩幅度增强;调宁蛋白和钙调蛋白结合蛋白基因表达量降低;细胞内游离钙的浓度增加。结论:半夏泻心汤防治反流性食管炎的机制可能是通过下调调宁蛋白和钙调蛋白结合蛋白基因表达,增加细胞内游离钙的浓度进而增加食管平滑肌收缩能力实现的。

Abstract:Objective:To explore the preventive and therapeutic mechanism of BX decoction on reflux esophagitis on the basis of esophageal movement.Methods:Sixty rats were randomly divided into 4 groups: sham operation group, model group,cisapride group and BX group.Gastrointestinal reflux models in rats were made.Physiograph was used to observe the contraction of esophageal muscle strips.An RT-PCR based assay was used to detect mRNA expression for calponin and caldesmon in esophageal smooth muscle.Intracellular free calcium was measured by laser scanning confocal microscopy.Results:Compared with that in model group,contractive range of esophageal muscle strips in BX group increased greatly,mRNA expression level for calponin and caldesmon decreased obviously and intracellular free calcium increased significantly.Conclusion: The preventive and therapeutic mechanism of BX decoction on reflux esophagitis may be due to enhancement of contraction of esophageal smooth muscle by downregulating of mRNA expression for calponin and caldesmon and increasing the intracellular free calcium.
