Abstract:Objective To preliminary study the effects and mechanism of coal-burning fluorosis on testosterone level in serum of rats.Methods SD rats taken as the experimental objects were randomly assorted into 4 groups which were control group,low-dose fluoride group,middle-dose fluoride group,and high-dose fluoride group.The exposed groups were fed with the fodder with different proportions of corn dried by burning coal coming from endemic fluorosis areas,to establish the animal model of coal-burning fluorosis.All rats were killed on the 180th day.Followed by examinations of dental fluorosis,fluorine in urine and the concentrations of testosterone in serum.Conventionally produced HE stained pathological sections of adrenal and testicular and testicular ultrathin sections,observed the structure changes of zona reticularis of adrenal gland and Leydig cell.Results The animal model of fluorosis was successfully estblished.Compared with the control groups,the level of serum testosterone declined in every exposed groups(P<0.01).With the increase in fluoride dose,declination was even more apparent(P<0.01).Observed by light microscopy,the zona reticularis of adrenal and Leydig cells were not obvious damaged.Observed by transmission electron microscopy,Leydig cells were damaged significantly.A large number of cytoplasmic lipofuscin granules,and even nuclear condensation,dissolution,cell rupture were observed.Conclusion The level of coal-burning fluorosis rats’ serum testosterone declined,Leydig cells’damage is the main reason.
