银杏叶提取物对犬低温体外循环下肺MMP-2和MMP-9 mRNA的影响

摘    要:目的探讨银杏叶提取物EGb(Ginkgo biloba Extract,EGb)对犬低温体外循环时肺组织MMP-2(matrix metalloproteinase-2,MMP-2)和MMP-9(matrix metalloproteinase-9,MMP-9)mRNA表达的影响。方法将14只健康杂种犬随机分为对照组(C组)和银杏叶提取物组(EGb组),每组7只。EGb组在麻醉后开胸之前给予银杏叶提取物8 mg/kg溶于0.9%生理盐水100 mL静脉滴注,C组以等量生理盐水静脉滴注。体外循环60 min,心脏复跳并循120 min后取肺组织用半定量RT-PCR检测基质金屈蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)mRNA的表达水平。结果与C组比较,EGb组肺组织MMP-2和MMP-9 mRNA的表达水平明显下调。结论银杏叶提取物可下调犬低温体外循环时肺MMP-2、MMMP-9 mRNA的表达。

Abstract:Objective The mRNA level of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in lung tissue was investigated about crossbred canines treated by ginkgo biloba extract undergoing CPB.Methods 14 dogs were divided into 2 group(n=7)contained control group and EGb group by Sequential method.The 8 mg/kg of EGb was infused via CVP before opening the thoracic cavity in EGb group,while the normal saline was used instead of EGb in control group.Blood samples were taken from internal jugular vein,broncho-alveolar lavage fluid was collected,and lung tissue was obtained 120 min after the aorta unclamping for determination of lung permeability index,W/D lung weight ratio,concents of MMP-2 and MMP-9 in lung tissue.Results EGb significantly decreased lung permeability index,W/D lung weight ratio,concents of MMP-2 and MMP-9 as compared with control group.Conclusion EGb can attenuate CPB-induced lung injury though decreasing the concents of RNA of MMP-2 and MMP-9 in lung.
