枳术颗粒对慢性萎缩性胃炎大鼠胃组织COX-2和VEGF表达的影响

摘    要:目的:观察枳术颗粒对慢性萎缩性胃炎(CAG)大鼠的疗效,并探讨其作用机制。方法:选取已建立CAG模型的大鼠,分为模型组,胃复春(0.86 g·kg-1)组,枳术颗粒高、中、低剂量(20,10,5 g·kg-1)组。用蛋白免疫印迹法(Western blot),免疫组化及实时荧光定量PCR(Real-time PCR)方法检测胃组织中环氧合酶-2(COX-2),血管内皮生长因子(VEGF)蛋白及mRNA的表达。结果:与正常组比较,模型组大鼠胃黏膜腺腔结构不完整,炎细胞浸润黏膜全层,体重增长率降低,COX-2,VEGF蛋白及mRNA表达显著增加(P0.01);与模型组比较,枳术颗粒高、中、低剂量组及胃复春组均显著增加CAG大鼠体重增长率(P0.01),下调大鼠胃组织COX-2,VEGF蛋白及mRNA表达(P0.01);与胃复春组比较,枳术颗粒高、中、低剂量组均显著增加CAG大鼠体重增长率(P0.01),高剂量组可下调大鼠胃组织COX-2,VEGF蛋白及mRNA表达,但无明显差异。结论:枳术颗粒可加快大鼠体重增长,使CAG大鼠胃黏膜组织得到改善和恢复。其作用机制可能为下调胃黏膜破坏因子COX-2,VEGF的表达,起到预防及治疗作用。

Abstract:Objective: To observe the effect of Zhizhu granules on the rat model with chronic atrophic gastritis (CAG). Method: The rat model with chronic atrophic gastritis was established, and divided into model group, positive control group (Weifuchun,0.86 g·kg-1), and Zhizhu granules high, medium and low-dose(20,10,5 g·kg-1)groups. The mRNA and protein expressions of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) were detected by Western blot, immunohistochemical (IHC) and Real-time PCR method. Result: Compared with normal group, model group showed incomplete gastric mucosa gland cavity structure, obvious inflammatory cell infiltration, significant decreasing in weight growth rate, and significant increase in COX-2 and VEGF protein and mRNA expressions (P<0.01). Compared with model group, Zhizhu granules high, medium and low-dose groups and Weifuchun group showed significant increase in CAG rats'' body weight growth rate (P<0.01), decrease in COX-2, VEGF protein and mRNA expressions in rat gastric tissues (P<0.01). Compared with Weifuchun group, Zhizhu granules high, medium and low-dose groups showed significant increase in CAG rats'' body weight growth rate (P<0.01), high-dose group could decrease the expression of COX-2, VEGF protein and mRNA expressions in rat gastric tissues, with no significant difference. Conclusion: Zhizhu granules can increase the growth of the body weight of rats, and also alleviate pathological morphology in stomach tissues. Zhizhu granules show the preventive and therapeutic effects by down-regulating the expressions of COX-2 and VEGF.
