摘    要:目的 利用大鼠胎鼠分离提取成骨细胞,观察含15%磷酸二氢钙+0.5%利塞膦酸骨水泥和1%利塞膦酸骨水泥对体外培养的大鼠成骨细胞OPG、RANKL和OC基因表达的作用.方法 实验分成空白对照组、15%磷酸二氢钙+0.5%利塞膦酸骨水泥浸提液组和1%利塞膦酸骨水泥浸提液组.实验组细胞培养基中加入相对应的磷硅酸钙骨水泥的PBS浸提液,浸提液以1:7与DMEM培养基混合作为细胞培养基;空白对照组成分以无菌PBS与DMEM培养基混合作为细胞培养基,每组6个平行,培养72 h后,提取细胞总RNA,应用RT·PCR法检测细胞中OPG、RANKL和OC的mRNA表达.结果 实验组成骨细胞中OPG和OC的mRNA表达较对照组的表达明显升高,RANKL的表达明显低于对照组,差异有统计学意义(P<0.05).结论 含15%磷酸二氢钙的0.5%利塞膦酸骨水泥和1%利塞膦酸骨水泥浸提液对SD大鼠胎鼠成骨细胞OPG和OC的表达具有促进作用,同时降低了RANKL的表达.

Effect of Bone Cement Containing the Risedrone on the Expression of OPG,RANKL and OC in SD Rat Osteoblasts

Abstract:Objective To isolate osteoblasts of from fetal rats and to investigate the effect of 0. 5% risedronate bone cement + 15% calcium dihydrogen phosphate and 1% risedronate bone cement on expression of OPG,RANKL and OC in cultured osteoblasts. Methods The experiment was divided into control group (PBS), leaching solution of 0. 5% risedronate +15% of calcium, dihydrogen phosphate and leaching solution from 1% risedronate. The PBS,leaching solutions were mixed with DMEM medium (1 : 7) ,and the isolated osteoblasts were cultured with the above mixed culture medium for 72 h. Total cell RNA was extracted and subjected to RT-PCR assay to examine the expression of OPG,RANKL and OC. Results The mRNA expressions of OC and OPG in two risedronate groups were significantly higher than the control group,while the RANKL expression was significantly lower than the control group (P〈0. 05). Conclusion 15% calcium dihydrogen phosphate + 0. 5% risedrone bone cement and 1% risedronate bone cement extract could promote rat fetal osteoblasts OPG and OC expression,while reduce the RANKL expression.
