应激活化蛋白激酶介导氯化镉致肾上腺皮质细胞凋亡作用探讨

摘    要:目的 了解氯化镉(CdCl2)对豚鼠肾上腺皮质细胞应激活化蛋白激酶(stress activated protein kinase,SAPK)活性的影响及与凋亡的可能关系。方法 以0.625、1.25和2.50mg/kg CdCl2整体腹腔注射染毒雄性豚鼠,分别于染毒1h和12h处死动物,取肾上腺皮质进行SAPK活性测定(免疫沉淀-蛋白印迹杂交-化学发光法)和电镜观察细胞凋讯。结果 整体染毒1h、SAPK活性随CdCl2染毒剂量的增加呈增加的趋势;染毒12h,各组SAPK活性变化不明显。电观察染毒1h肾上腺皮质细胞未见凋亡征象;但染毒12h可见早期细胞凋亡。结论 SAPK活化在CdCl2诱导肾上腺皮质细胞凋亡过程中可能起重要作用。

Abstract:Objective To observe the apoptosis of adrenocortical cells in guinea pig induced by cadmium chloride (CdCl2) and explore its possible correlation with alteration of SAPK (stress activated protein kinase) activity. Methods 40 male guinea pigs were randomly distributed into four groups which were administered with 0 (control), 0.625, 1.25 and 2.50 mg/kg CdCl2 by intraperitoneal injection respectively. The adrenal cortexs were collected after 1 hour and 12 hours of injection. The parameters including: ①detection of apoptosis with electronic microscope (EM). ②assays for SAPK activity. The adrenal cortexs were homogenized and the SAPK activities were measured with immuno-precipitation & chemiluminescence assay.Results The activities of SAPK were elevated as the doses of CdCl2 increased at 1 hour, but no remarkable alteration of SAPK activity of early apoptotic features could be observed by EM when guinea pigs were introperitoneally injected with 1.25 mg/kg and 2.50mg/kg CdCl2 at 12 hours. Conclusion It showed that the stress response and SAPK activation occurred earlier than apoptosis happened and suggested that SAPK might play an important role in apoptosis of adrenal cortex induced by CdCl2.
