脉冲激光对诱发型2型糖尿病大鼠神经病理性疼痛的影响研究

摘    要:目的:研究低强度脉冲激光对链脲佐菌素(streptozotocin,STZ)联合高脂高糖膳食诱导的2型糖尿病(type 2 diabetes,T2D)大鼠神经病理性疼痛的治疗效果。方法:使用完全随机分组法将SD大鼠分为对照组(Control组)、T2D大鼠组(T2D组)、T2D大鼠激光治疗组(T2D+laser组)3组,每组10只。对T2D+laser组大鼠施加激光治疗8周,每天3次,每次50 s,Control组和T2D组予以假暴露干预。检测并比较各组大鼠的足底机械痛阈值、足底热痛阈值以及脊髓白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和神经生长因子(NGF)的基因表达水平。使用SPSS 22.0软件进行统计学分析。结果:T2D组大鼠足底机械痛阈值和热痛阈值显著低于Control组(P<0.01),脊髓中IL-1β、TNF-α和IL-6的基因表达水平显著高于Control组(P<0.01),NGF基因表达水平显著低于Control组(P<0.01)。低强度脉冲激光治疗的第2、4、6、8周显著提高了T2D大鼠足底机械痛阈值和热痛阈值(P<0.01),显著降低了T2D大鼠脊髓中IL-1β、TNF-α和IL-6的基因表达水平(P<0.01),并显著提高了NGF的基因表达水平(P<0.05)。结论:低强度脉冲激光疗法能够显著缓解诱发型T2D大鼠神经病理性疼痛症状。

Abstract:Objective To study the therapeutic effect of low-intensity pulsed laser on neuropathic pain induced by streptozo-tocin(STZ)combined with high-fat and high-sugar diet in type 2 diabetic rats.Methods Totally 30 SD rats were divided into 3 groups of 10 rats each:control(control group),T2D rats(T2D group)and T2D rats with laser treatment(T2D+laser group)using a completely randomized grouping method.The rats in the T2D+laser group were treated with laser for 8 weeks,three times a day for 50 s.The control and T2D groups were given sham exposure.The paw mechanical pain thresholds,paw thermal pain thresholds and the gene expression levels of spinal interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and nerve growth factor(NGF)were measured and compared between the rats of the groups.SPSS 22.0 software was used for statistical analysis.Results The rats in the T2D group exhibited significantly lower paw mechanical pain threshold and thermal pain threshold than those in the control group(P<0.01),higher spinal gene expression levels of IL-1β,TNF-αand IL-6 while lower NGF expression than those in the control group(P<0.01).Low-intensity pulsed laser treatment significantly inhibited the decrease of paw mechanical pain threshold and thermal pain threshold in T2D rats at 2,4,6,and 8 weeks after treatment(P<0.01).Moreover,pulsed laser treatment significantly reduced the spinal gene expression levels of IL-1β,TNF-αand IL-6 in T2D rats(P<0.01),and also significantly upregulated the spinal NGF expression level(P<0.05).Conclusion Low-intensity pulse laser therapy can significantly attenuate the neuropathic pain symptoms in induced type 2 diabetic rats.
