Abstract:Objective To investigate the effects of CXCL4 gene interference on malignant proliferation and epithelial mesenchymal transformation of GBC-SD cells.Methods shRNA CXCL4 vector was constructed and transfected into GBC-SD cells.The cells were randomly divided into three groups:Control group,shRNA-NC group and CXCL4-shRNA1 group.Cell proliferation was detected by EDU staining.Flow cytometry was used to detect apoptosis.Morphological changes of epithelial mesenchymal transformation were observed by fluorescence microscopy.Western blot was used to detect the expressions of Ki67,Survivin,Bax,Bcl-2,E-cadherin and N-Cadherin.Immunofluorescence was used to detect Vimentin.Results Compared with the Control group,the number of EDU positive cells and the levels of Ki67,Survivin,and N-cadherin in the CXCL4-shRNA1 group were decreased,and the apoptosis rate,Bax/Bcl-2 ratio,E-cadherin level,the inhibition of epithelial stromal transformation,and the level of Vimentin in the CXCL4-shRNA1 group were all significantly decreased(P<0.05).Conclusion The CXCL4 gene interfered inhibits the malignant proliferation,epithelial-mesenchymal transformation and promotes apoptosis of GBC-SD cells.
