Abstract:Objective To investigate the effect of basic fibroblast growth factor(bFGF) and its monoclonal antibody(bFGF-Mab) with different concentrations on angiogenesis and growth of subcutaneously transplanted human invasive pituitary adenoma in nude mice and the possible mechanism.Methods Fifty mouse models of subcutaneously transplanted human invasive pituitary adenoma were established and were randomly divided into 5 groups including a bFGF group,a bFGF-Mab 30 mg/ml group,a bFGF-Mab 20 mg/ml group,a bFGF-Mab 10 mg/ml group and a normal saline group(10 mice in each group).The mice were treated for 8 weeks.Tumor volumes were measured,and growth curves of transplanted tumors were drawn every week.Immunohistochemistry was applied to detect CD31 expression to calculate tumor microvessel density(MVD).Results As compared with the control group and the bFGF group,the growth and angiogenesis of transplanted tumor in each bFGF-MAb group were significantly inhibited(P<0.01);the inhibition rate was higher(55.71%) in the bFGF-Mab 30 mg/ml group than that in the other two bFGF-Mab groups.MVD expression in each bFGF-MAb group was all decreased;the decrease was significant in the bFGF-Mab 30 μg/ml group(3.36±1.26) as compared with other two bFGF-MAb groups(P<0.01).Conclusion bFGF-Mab has a strong inhibitory effect on the nude mice of subcutaneously transplanted human invasive pituitary adenoma through the inhibition of tumor proliferation,invasion and angiogenesis.
