磷酸钙人工骨结合骨髓基质干细胞移植修复骨缺损的实验研究

摘    要:目的制备磷酸钙人工骨(calcium phosphats artifieal bone, CPC)与骨髓基质干细胞(bone mesenchymal stem cells, BMSCs)复合物,通过动物实验研究其对骨缺损的修复作用。方法分离、扩增兔的BMSCs,并以CPC为载体制备cPc/BMSCs复合物,植入兔桡骨15mm骨缺损处。于术后不同时间处死动物,通过X线摄片、组织学染色分析和放射性核素监测,观察新骨形成和材料降解情况。同时以单纯的CPC及空白对照组作为对照研究,评价CPC/BMSCs复合物对骨缺损的修复能力。结果CPC/BMSCs复合物可再生新骨组织并完好地修复桡骨缺损,且其修复能力明显优于单纯的CPC。结论以CPC为载体的自体骨髓细胞移植能有效的修复骨缺损。

Transfer of calcium phosphate artificial bone combined with mesenchymal stem cells for repair of segmental bone defects:an experimental study
Abstract:Objective To construct composite of calcium phosphate cement (CPC) and bone mesenchymal stem cells (BMSCs) and to evaluate its function in repairing the bone defect.Methods BMCSs was isolated and amplified. CPC/ BMSCs complex was formed using calcium phosphate cement as carrier. The bone defects of 15mm in length in the radius were implanted with CPC/ BMSCs composite. The specimens were harvested separately at various interval. The formation of new bone and the degradation process of the material were observed under a series of examinations, including radiography, histomorphology and radionuclide bone imaging. The results of CPC/BMSCs were compared with those in simple CPC and control group for the evaluation of the effect of the composite of CPC/BMSCs on repairing bone defect. Results CPC/ BMSCs composite can form new bone and repair bone defect. The repair ability was superior to that in simple CPC group. Conclusion The composite of bone marrow stem cells using calcium phosphate cement as carrier can repair the bone defects efficiently.
