二次包埋法手工制作病理学教学组织芯片

摘    要:目的探讨手工制作教学组织芯片方法的可行性。方法利用组织芯片制备系统,通过二次包埋法,手工制作病理教学组织芯片,经HE染色,光镜下检测组织芯片制作质量。采用四川省病理技术专业委员会对石蜡切片HE制片质量评定得分标准对切片进行评分,并与传统教学组织切片比较。结果组织微阵列(TMA)蜡块中的组织芯点排列整齐,与受体蜡块融合紧密;组织芯片切片基本完整,厚薄均匀,经HE染色后组织形态可利用率为94%~100%,切片质量评分优秀率79%,良好率10%。二次包埋法手工制作的教学组织芯2片与传统组织切片制作方法制作的教学组织切片质量无明显差异性(χ=3.073,P0.05)。结论二次包埋法手工制作教学组织芯片方法简便易行,经济实用低消耗,具有高效快速染色均一性好、自身内对照和可比性强的特点,可在病理教学中推广应用。

Abstract:Objective To explore the feasibility of hand-making the teaching tissue microarray. Methods Using tissue microarray preparation system, hand-crafted pathology teaching tissue microarray was made by double embedding method and stained with HE to evaluate the production quality by light microscopy. According to Paraffin-embedded sections HE quality assessment score standards of Sichuan Pathological Technical Committee, the sections were assigned into numerical scores and compared with the traditional teaching tissue sections. Results Core points of the paraffin tissue microarray(TMA) were arranged in neat rows and in close integration with the receptor paraffin block, with basic integrity, uniformity thickness. Stained with HE, the utilization rate of tissue morphology was 94%~100%, the excellent rate of sections quality rating was 79%. There was no significant difference for the quality of teaching tissue sections between double embedding method and traditional method (χ2=3.073, P0.05). Conclusion Double embedding method is a simple, economical and practical with highly efficient and nice dyeing uniformity, which can be widely applied in pathology teaching.
