EGCG对舌鳞癌Tca8113细胞增殖与凋亡的实验研究

摘    要:目的:研究表没食子儿茶素没食子酸酯(EGCG)对体外培养的人舌鳞癌Tca8113细胞增殖抑制及诱导凋亡的作用。方法:采用CCK-8法检测不同浓度的EGCG(0、10、20、40、80mg/L)对Tca8113细胞的体外增殖抑制作用;采用流式细胞术及DAPI荧光染色法检测EGCG对Tca8113细胞凋亡的影响。结果:CCK-8法检测显示EGCG对Tca8113细胞有明显增殖抑制作用并呈现时间及浓度依赖性(P<0.01);流式细胞仪结果显示,经10mg/L EGCG处理的Tca8113细胞凋亡率还不能认为与对照组有区别,其余EGCG处理组细胞凋亡率均高于对照组并呈现时间及浓度依赖性(P<0.01);DAPI染色在免疫荧光显微镜下可见典型的细胞核皱缩及凋亡小体等。结论:EGCG对Tca8113细胞具有明显的增殖抑制及诱导凋亡作用,且呈现时间及浓度依赖性。

Effects of EGCG on the Proliferation and Apoptosis of Human Oral Squamous Cell Carcinoma Tea8113 Cell line

Abstract:Objective: To investigate the effects of Epigallocatechin-3-gallate(EGCG) on the proliferation and apoptosis of human oral squamous cell carcinoma Tca8113 cell line in vitro.Methods: The proliferation of the Tca8113 cells treated by different concentrations of EGCG(0,10,20,40,80μg/mL) was assayed by Cell Counting Kit-8(CCK-8) method.The cell apoptosis was detected by Flow cytometric analysis,and the morphological alterations of the cancer cells were observed under immunofluorescence microscopes.Results: In comparison to the control,the results of the CCK-8 method display that all the different concentrations of EGCG were able to inhibit the growth of the cell lines,and it increased in a time-and-dose-dependent manner(P<0.01).The apoptosis rate is like this(P<0.01) except in the 10μg/ml EGCG group which was detected by Flow cytometric analysis.After being treated with EGCG,the cell nucleus shrink typically and apoptotic body were seen under immune fluorescence microscopy with DAPI dyeing.Conclusion: EGCG could effectively inhibit the proliferation and induce apoptosis of Tca8113 cell lines in a time and dose dependence.
