透明质酸对体外培养颞下颌关节滑膜成纤维细胞粘附与增殖性能的影响

摘    要:目的:研究不同浓度的高分子量透明质酸(hyaluronic acid,HA)对体外培养的滑膜成纤维细胞的粘附与增殖的影响。方法:分离兔颞下颌关节滑膜组织,组织块法进行滑膜成纤维细胞的原代及传代培养。采用不同浓度HA(0%,0.01%,0.05%,0.1%,0.5%,1%)作用于滑膜成纤维细胞,观察对其粘附与增殖性能的影响,通过单因素方差分析(One-way ANOVA)来评价结果的统计差异性。结果:0.05%和0.1%的HA对细胞粘附和增殖的促进作用明显(P〈0.05),0.5%和1%的HA对细胞的粘附与增殖起抑制作用(P〈0.05),低于0.05%的HA对细胞粘附与增殖无明显促进作用。0.05%和0.1%HA组对增殖的促进作用随时间而增强。结论:低浓度的HA对细胞粘附和增殖的无明显促进作用,而高浓度的HA会抑制细胞的粘附和增殖。

Abstract:Objective:To evaluate the effects of high molecular weight hyaluronic acid(HA)on adhesion and proliferation of synovial fibroblasts from the temporomandibular joint.Methods:The synovial tissues were harvested from the rabbit TMJ,and the synovial fibroblasts were separated and cultured by tissue block method.Different concentrations of HA(0%,0.01%,0.05%,0.1%,0.5%,1%)were added to the culture system.Effects of HA on adhesion and proliferation of the synovial fibroblasts were observed.The results were analyzed by one-way ANOVA.Results:The adhesion and proliferation of the synovial fibroblasts was significantly promoted by HA in the 0.05%and 0.1% group(P<0.05).The adhesion and proliferation synovial fibroblasts were inhibited when the HA concentration is higher than 0.1%(P<0.05).There was no significant difference when HA concentration is below 0.05%.Conclusion:HA,with a concentration between 0.05%and 0.1%,may promote the adhesion and proliferation of synovial fibroblasts from the temporomandibular joint.
