纳米银干预牙本质表面粪肠球菌粘附的实验研究

摘    要:目的:研究纳米银对牙本质表面粪肠球菌粘附的干预作用。方法:制备牙本质片42个,随机均分成3组,分别置于0.1%纳米银溶液、1.313%次氯酸钠溶液和生理盐水内浸泡30min后,于200μL粪肠球菌悬液中培养1h,用扫描电镜观察牙本质表面形态和粪肠球菌粘附情况,荧光显微镜分析牙本质表面粘附的粪肠球菌数量并拍照记录,Image pro plus 6.0软件计算粪肠球菌总菌数、活菌数和活菌百分比。结果:0.1%纳米银溶液组和生理盐水组牙本质表面形态清晰无破坏,1.313%次氯酸钠溶液组牙本质溶融坍塌,胶原溶解破坏。牙本质经0.1%纳米银溶液处理后,其表面粘附的粪肠球菌总菌数、活菌数和活菌百分比均低于1.313%次氯酸钠溶液组和生理盐水组(P﹤0.01)。结论:0.1%纳米银溶液可有效减少粪肠球菌对牙本质的粘附,对牙本质表面形态无明显破坏。

Effect of Nano-silver on the Adhesion of Enterococcus Faecalis to Dentin Surface

Abstract:Objective: To evaluate the influence of nano-silver on the adhesion of Enterococcus faecalis to dentin surface.Methods: 42 dentin slices were randomly divided into 3 groups of 14 specimens,and 3 groups of dentin slices were correspondingly immersed in 0.1% nano-silver solution,1.313% sodium hypochlorite solution and 0.9% physiological saline for 30min,and then each dentin slice was inoculated with 200μL of E.faecalis suspension for 1h.3 specimens of each group were used for scaning electron microscope observation of dentin surface morphologies and adhesion situations of E.faecalis to dentin slices,and 11 specimens were stained with BacLight for examining adhesion situations and taking pictures by fluorescence microscope.The viable count,total bacteria count and percentage of viable cells of E.faecalis adhered to dentin slice were analyzed by Image pro plus 6.0.Results: The surface morphologies of dentin slices treated by 0.1% nano-silver solution and 0.9% physiological saline displayed no destruction,while dentin slices treated by 1.313% sodium hypochlorite solution were destroyed with dissolution of collagen.The viable count,total bacteria count and percentage of viable cells of E.faecalis adhered to dentin slices treated by 0.1% nano-silver solution were lower than 1.313% sodium hypochlorite group and 0.9% physiological saline group(P﹤0.01).Conclusion: 0.1% nano-silver solution could reduce the adhesion of E.faecalis to dentin and have no influence on the surface morphology of dentin.
