米诺环素对成年大鼠下颌骨成骨细胞内外金黄色葡萄球菌的作用

摘    要:目的:探讨米诺环素对金黄色葡萄球菌感染的成年大鼠下颌骨成骨细胞的抗菌作用。方法:采用金黄色葡萄球菌ATCC 25923感染体外培养的成年大鼠下颌骨成骨细胞,被感染的成骨细胞部分经庆大霉素处理后加入米诺环素,另一部分直接加入米诺环素,溶解细胞后计算细胞内外的菌落数。结果:经庆大霉素杀灭细胞外细菌后,细胞内细菌在米诺环素的作用下,随着时间的延长细菌数量减少,24h后基本抑制细胞内菌;随着米诺环素直接作用感染细胞的时间延长细胞内外的细菌均减少,3h细胞外细菌基本抑制,24h后细胞内的细菌也基本完全抑制。结论:米诺环素能够抑制成骨细胞内外的金黄色葡萄球菌,具有时间依赖性,为种植体给药系统的临床应用奠定实验基础。

Antibacterial Activity of Minocycline against Staphylococcus Aureus inside and outside the Mature Rat Mandibular Osteoblasts

Abstract:Objective: To evaluate the antibacterial activity of minocycline in a Staphylococcus aureus-mature rat alveolar osteoblast model.Methods: Mature rat alveolar osteoblasts were infected by Staphylococcus aureus ATCC 25923.There were two groups in infected cells.After killing extracellular Staphylococcus aureus with gentamicin,minocycline was added in one group.Minocycline was added directly in another group.After dealing with minocycline,infected osteoblasts of two groups were both lysed and cultured for extracellular and intracellular bacterium colony forming units(CFU).Results: After the extracellular bacteria were killed by gentamicin,the intracellular bacteria were reduced gradually with the prolongation of minocycline effect and were almost inhibited completely after 24 hours.The intracellular and extracellular bacteria were reduced simultaneously with the prolongation of minocycline effect after Staphylococcus aureus invading osteoblast.And extracellular bacteria were almost inhibited after 3 hours,almost all intracellular bacteria were not present after 24 hours.Conclusion: Minocyclines can inhibit the Staphylococcus aureus inside and outside the osteoblasts with time dependence supporting its potential clinical utilization through special implant administer system.
