骨桥蛋白与口腔鳞状细胞癌细胞生物学特性关系的实验研究

摘    要:目的 初步探讨骨桥蛋白(osteopontin,OPN)与口腔鳞状细胞癌(鳞癌)细胞生物学特性关系。方法 体外培养口腔鳞癌细胞株KB和FaDu细胞,MTT法比较2种细胞的增殖活性,基质胶侵袭实验和划痕实验检测比较2种口腔癌细胞的侵袭和迁移能力的差异,免疫组化SP法检测细胞中OPN的表达。结果 KB细胞的增殖、侵袭和迁移能力均高于FaDu细胞,两种细胞中均有OPN的表达,KB的表达量高于FaDu。结论 口腔鳞癌细胞株KB和FaDu细胞生物学特性的差异可能与其OPN表达差异有关,高表达OPN的KB细胞株可以作为体外研究口腔鳞癌中OPN基因作用的实用细胞株。

Abstract:Objective This study was to assess the relationship of osteopontin(OPN) and biological characteristics of two oral squamous cell carcinoma(OSCC) cell lines.Methods The proliferation,invasiveness and mobility abilities of KB and FaDu cell lines were determined and compared with MTT method,matrigel invasion assays and wound healing respectively.OPN protein expression was detected by immunohistochemical streptavidin/peroxidase(SP) in oral squamous cell carcinoma cell lines KB and FaDu.Results The proliferation,invasiveness and mobility abilities of KB were much stronger than those of FaDu.OPN both expressed in KB was and FaDu,and the expression in KB was higher than that in FaDu.Conclusions The different biological characteristics of two oral squamous cell carcinoma cell lines may be due to of different levels of OPN expression.High expression of OPN in KB cell line may make KB cell line a useful cell line for in vitro studies designed to clarify the role of OPN in OSCC.
