摘 要: | 目的:建立维甲酸诱导雌性大鼠骨质疏松模型。方法:20只4月龄雌性Wistar大鼠随机均分为2组:对照组和实验组,分别给予维甲酸(70mg/kg/次)和生理盐水灌胃4周,末次给药后测量血清Ca、P、雌二醇(E2)、碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶(TRAP)含量,双能X线骨密度仪活体测量腰椎、胫骨以及股骨密度,动物处死后取胫骨行组织学观察。结果:停药后血清学指标测定,实验组血清Ca、P与对照组比较差异无显著性,血清E2明显低于对照组(P<0.01),血清ALP、TRAP明显高于对照组(P<0.01)。腰椎、胫骨及股骨密度检测显示实验组骨密度明显低于对照组(P<0.05),组织学观察亦支持骨密度测量结果。结论:根据血清学指标、骨密度检测及组织学观察结果,表明维甲酸诱导大鼠骨质疏松模型制作成功。

Abstract: | Objective: To establish osteoporosis rat model induced by retinoic acid.Methods: Twenty female Wistar rats of 4 months old were selected and randomly divided into 2 groups of 10 rats each.Rats in experimental group were treated with retinoic acid at the dose of 70mg/kg for 4 weeks,while the control group was given the same dose of distilled water.Serum biochemical indexes of Ca,P,E2,ALP and TRAP were determined for the 2 groups at the end of the last dose.Densitometry of lumbar,tibiae and femur was simultaneously performed by dual-energy X-ray absorptiometry.Histological changes in tibiae were also observed after animal sacrifice.Results: Compared with the control group,the serum biochemical indexes of Ca,P were no significant difference,the serum biochemical indexes of E2 were significantly lower(P<0.01),the serum biochemical indexes of ALP,TRAP were significantly higher(P<0.01),and the densitometry of lumbar,tibiae and femur was significantly lower in the experimental group(P<0.05).The observation under microscope supported the results of bone density measurement.Conclusion: The results of serum biochemical indexes,the densitometry of lumbar,tibiae and femur,the histological appearance indicated that the animal model of osteoporosis with rats by retinoic acid was established. |
