肠道菌群总DNA提取方法的研究

[目的]通过实验得出提取DNA的最佳方法,并对样品处理等实验条件进行优化,为PCR检测中模板DNA的制备提供实验依据。[方法]一.样品处理;二.DNA提取:1.醋酸钠-乙醇沉淀法;2.水煮法;3.水煮法-试剂盒法;4.试剂盒法;5.CTAB法;三.DNA的检测。[结果]实验效果最佳的DNA提取方法是试剂盒法,所得到的结果(包括DNA纯度和浓度)是几种提取方法中最好的;水煮法-试剂盒法结果较试剂盒法相近,但操作较烦琐;醋酸钠-乙醇沉淀法、CTAB法和水煮法得到的DNA中含有较多的蛋白质、RNA等杂质。[结论]试剂盒法是提取DNA最佳方法。

STUDY ON THE DNA EXTRACTION METHOD OF THE INTESTINAL FLORA

[Objective]To get the best method of DNA extraction and optimize the condition of exprimentation just like the dispoal of sample and so on by experiment,and provide experimental reference for PCR.[Methods]1.Samples dispose;2.DNA extraction:1.Sodium acetate-ethanol precipitation;2.Boiled;3.Boiled Act-kit;4.Kit and 5.CTAB.[Results]The best method for DNA extraction was kit.The result of Boiled Act-kit was close to kit,but there were lots of process.The DNA extract by Sodium acetate-ethanol precipitation and boiled had more impurity just like RNA and protein and so on.[Conclusion]Kit is the best way to extract DNA.
