黄芪多糖对鼻咽癌CNE-2细胞增殖抑制作用的研究

摘    要:目的]研究黄芪多糖(APS)对人鼻咽癌CNE-2细胞增殖的影响。方法]设不同浓度APS(12.5,25.0,50.0,100.0μg/ml)、二甲基亚砜2‰为空白对照组,顺铂2μg/ml为顺铂组。应用MTT法检测APS对鼻咽癌CNE-2细胞的增殖抑制作用。结果]与空白对照组比较,不同浓度的APS组在12、24、48、72h作用时吸光度均表现为上升的趋势,在48h时达到最大值,随后下降。不同浓度的APS组经不同的作用时间后,其吸光度与空白对照组比较差异均有统计学意义(P﹤0.05)。与顺铂组比较,12.5、25.0μg/mlAPS作用鼻咽癌CNE-2细胞12h时细胞的增殖没有明显的抑制作用(P﹥0.05),但作用24、48、72h时对鼻咽癌CNE-2细胞的增殖有明显的抑制作用(P﹤0.05);50.0、100.0μg/mlAPS在不同的作用时间对鼻咽癌CNE-2细胞的增殖均有明显的抑制作用(P﹤0.05)。结论]APS可以明显地抑制鼻咽癌CEN-2细胞的生长,且存在剂量-时间的关系。

Abstract:Objective]To investigate the antir proliferation effect of astragalus polysaccharides(APS)on human nasopharyngeal carcinoma CNE-2 cells,and explore the molecular mechanism of anti-NPC.Methods]This study set up APS different concentrations groups(12.5,25.0,50.0 and 100μg/ml),2‰ DMSO blank control group and 2μg/ml cisplatin group.The antir proliferation effect of APS on human nasopharyngeal carcinoma CNE-2 cells was detected by MTT method.Results]Compared with the blank control group,the OD value of APS different concentrations group in 12,24,48 and 72 h showed an absorbance rise,at 48 h OD value the reached the maximum and then decreased.The OD value of APS different concentrations group in different culture time compared with the blank control group were significantly different(P﹤0.05).Compared with cisplatin group,there was no significant antir proliferation effect of 12.5μg/ml and 25.0 μg/ml APS concentrations group in 12h,but there was significant antir proliferation effect in 24,48 and 72h(P﹤0.05).The was significant antir proliferation effect in the 50.0μg/ml and 100.0 μg/ml APS concentrations group through different culture time(P﹤0.05).Conclusion]The APS is capable of significantly inhibitiing proliferation of CNE-2 cells in vitro,and the result of inhibiting proliferation rate shows a significant dose-time relationship.
