不同提取方法对虎杖中蒽醌类成分及其抗氧化活性影响的研究

摘    要:目的研究不同提取方法对虎杖蒽醌类成分及其抗氧化作用的影响。方法采用回流、索氏、微波及超声四种提取方法,测定四种提取液中的游离蒽醌和总蒽醌的含量;采用DPPH,FRAP两种方法测定其抗氧化活性,并分析蒽醌类成分和抗氧化活性之间的关系。结果在对游离蒽醌的测定中以索氏法为最高(0.53%),其次为超声(0.45%),各种方法之间相比差异显著(P<0.05);在对总蒽醌的测定中,结果如下:索氏(1.16%)>超声(1.15%)>微波(1.04%)>回流(0.80%);抗氧化活性测定发现,以回流液的抗氧化能力最强,其次是微波,二者与超声和索氏的相比有显著性差异(P<0.05);相关性分析发现,蒽醌的含量与抗氧化活性呈显著的负相关(P<0.01)。结论不同提取方法影响虎杖蒽醌类成分和抗氧化活性,蒽醌类成分和抗氧化活性之间具有负相关的关系。

Abstract:Objective To explore the effect of different extraction methods on the content of anthroquinone compound and its antioxidative activity in Polygonum cuspidatum Sieb.et Zucc..Methods Anthroquinone compound was extracted respectively by reflux method,Soxhlet extraction method ultrasound wave method and microwave-assisted extraction.The free and total anthroquinone content and its antioxidative activity was determined by spectrophotometry.The correlation between anthroquinone compound and antioxidative activity was analyzed.Results There were significant differences in the extraction content of free anthroquinone content among the four extraction method stated(P<0.05),Soxhlet extraction method was the highest(0.53%),ultrasound wave method was better(0.45%).In the determination of total anthroquinone,the result was:Soxhlet extraction method(1.16%)> ultrasound wave method(1.15%)microwave-assisted>reflux method > extraction.The reflux method was better than microwave-assisted extraction,compared with Soxhlet extraction method and ultrasound wave method.There were significant differences in the determination of its antioxidant activity(P<0.05).There was significant negative correlation between anthroquinone compound and its antioxidative activity (P<0.01).Conclusion Anthroquinone compound and antioxidative activity in Polygonum cuspidatum Sieb.et Zucc.was affected by different extracting methods,there was negative correlation between anthroquinone compound and antioxidative activity.
