缺氧对大鼠海马神经元生物学功能的影响

摘    要:为进一步探讨缺氧缺血如何诱导海马神经元损伤直至凋亡,明确缺氧对海马神经元生物学功能的影响及其相关机制,从而为缺血缺氧性脑病的防治提供实验依据,本研究以19d的SD胎鼠脑作为取材对象,经过条件培养基纯化建立海马神经元原代培养体系,通过MTT法、TUNEL法、免疫荧光、化学发光等方法检测海马神经元生长增殖活力、凋亡、Caspase-3活性等指标。结果显示缺氧可抑制SD大鼠海马神经元的生长增殖活力,并伴随着神经元凋亡和Caspase-3活性增高,且随着缺氧时间的增长程度加重。以上结果说明,缺氧通过诱导细胞凋亡导致SD大鼠海马神经元的损害,Caspase-3活性的激活可能是其重要的通路之一。

Abstract:In order to afford the experiment basis for ischemic encephalopathy prevention,the influence and mechanism of hypoxia on biological function of hippocampal neurons were discussed by investigating the hippocampal neurons damage and apoptosis induced by hypoxia. The 19 d SD embryo rat brain was taken as a material object and established the hippocampal neuron primary culture system by condition culture medium. The proliferation,apoptosis and Caspase-3 activity of the hippocampal neurons were detected through MTT,TUNEL,immunofluorescence and chemiluminescence. The results showed that hypoxia suppressed the neuron's growth multiplication vigor and increased the apoptosis and Caspase-3 activity. The suppression of growth level increased with the hypoxia time. Taken together,the results indicte that hypoxia induces apoptosis and leads to the SD rat hippocampal neurons damage,the activation of Caspase-3 activity may be one of its important pathway.
