胰岛素样生长因子在PCOS病人卵泡液及颗粒细胞中的表达

摘    要:目的:研究超促排卵周期中,胰岛素样生长因子Ⅰ及胰岛素样生长因子Ⅱ,在多囊卵巢综合征病人卵泡液的浓度和在颗粒细胞中的表达,探讨二者在多囊卵巢的病理改变,并预测在超促排卵周期中与卵巢过度刺激综合征的关系.方法:用酶联免疫吸附法检测取卵日外周血清、卵泡液中胰岛素样生长因子Ⅰ、胰岛素样生长因子Ⅱ的浓度,用免疫组化法检测卵泡颗粒细胞中胰岛素样生长因子Ⅰ、胰岛素样生长因子Ⅱ的表达.结果:胰岛素样生长因子Ⅱ在多囊卵巢综合征病人卵泡液中的浓度较非多囊卵巢综合征的浓度高,胰岛素样生长因子Ⅰ在两组之间的浓度无差异.胰岛素样生长因子Ⅱ在多囊卵巢综合征病人卵泡颗粒细胞中的表达强于非多囊卵巢综合征病人.结论:胰岛素样生长因子Ⅰ、Ⅱ在超促排卵周期中的产生受其它原因影响可能有所改变,胰岛素样生长因子Ⅱ在超促排卵周期中增高可能通过结合胰岛素样生长因子Ⅰ受体,导致卵巢多囊改变,多卵泡发育,易发生卵巢过度刺激综合征.

THE EXPRESSION OF IAF IN FOLLICULAR FLUID AND GRANULOSA CELL IN PAILENT WITH PCOS

Abstract:Objective:To study the levels of insulin-like growth factor Ⅰ(IGF-Ⅰ),insulin-like growth factor Ⅱ(IGF-Ⅱ)in serum and follicular fluid and their expression in granulosa cell in control ovary hyperstimulation(COH)cycle,the two factors were measured in polycystic ovary syndrome(PCOS)and control groups.Which would help to explain further the pathological changes of polycystic ovarian syndrome and potential Relationship with oarian hyperstimulation syndrome.Methods:Using method to detect the concentration of IGF-Ⅰ,IGF-Ⅱin serum and follicular liquid on the day of Ocyte retrieval,using immunohistochemical method to detect the expression of IGF-1 and IGF-Ⅱin he Granulosa cells.Results:The concentration of IGF-Ⅱin follicular liquid was higher in PCOS group than control groups.The expression of insulin-like growth factor II in follicular granulosa cells of patients with polycystic ovary syndrome was stronger than in patients with non-polycystic ovary syndrome.Conclusion:IGF-Ⅰ,IGF-Ⅱmay be affected by other reasons in COH cycle.By combining IGF-1 receptors,IGF-Ⅱmay contribute to ovarian polycystic change and the Multiple follicular growingin COH)cycle.And contribute to the happening of ovarian hyperstimulation syndrome.
