c—fos反义寡核苷酸对谷氨酸诱导PC12及海马神经元凋亡的影响

目的：探讨反义寡核苷酸c-fos对谷氨酸诱导培养的海马神经元及PC12凋亡的影响。方法：应用TUNEL（Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling)法和免疫细胞学方法检测了c-fos反义寡核苷酸对谷氨酸诱导原代培养的海马神经元及PC12细胞凋亡以及对凋亡蛋白Bax表达的影响。结果：与对照组相比，c-fos反义寡核苷酸谷氨酸谷氨酸诱导原供培养的海马神经元及PC12细胞的凋亡率减少和凋亡蛋白Bax表达量下降，差异有显著意义，结论：c-fos反义寡核苷酸对谷氨酸诱导的细胞凋亡有保护作用。

The effect of c-fos antisense oligonucleotide on apoptosis of glutamate-induced hippocampal neurons and PC12 cell line

Objective To probe the effect of c fos antisense oligonucleotide on glutamate induced hippocampal neurons and PC12 cell line. Methods Using TUNEL assay, we detected the apopotosis of glutamate induced hippocampal neurons and PC12 cells and investigated Bax expression on both cells by means of immunocytochemistry. Results The ratio of cells with property of appoptosis to normal cells is lower in the groups treated with c fos antisense oligonucleotide plus Glutamate than that of Glutamate groups ( P < 0.01 ). The average optic density in the group treated with c fos antisense oligonucleotide plus Glutamate is lower than that of the glutamate group, the difference is statistically significant ( P < 0.01 ). Conclusion The results suggest that c fos antisense oligonucleotide have anti apoptotic effect on glutamate induced hippocampal neurons and PC12 cells in culture.
