左旋四氢巴马汀在大鼠全脑缺血再灌注时对脑组织钙含量及脑细胞凋亡的影响

摘    要:目的 通过观察脑缺血再灌注时脑组织钙含量及脑细胞凋亡的变化 ,探讨左旋四氢巴马汀在全脑缺血再灌注损伤时的作用机制。方法 本实验在建立大鼠急性全脑缺血再灌注损伤模型的基础上 ,用原子分光光度仪检测脑组织钙含量 ,用流式细胞仪检测脑细胞凋亡。结果 脑缺血再灌注 12h钙含量较假手术组增高 (P <0 0 1) ,脑细胞凋亡数亦增加(P <0 0 1) ;2 4h钙含量进一步增高 (P <0 0 1) ,凋亡细胞数亦进一步增加 (P <0 0 1) ;左旋四氢巴马汀在脑缺血再灌注 12h和 2 4h均能抑制脑组织钙含量的增高 (P <0 0 1)并减少脑细胞凋亡数 (P <0 0 1)。各组脑组织钙含量与脑细胞凋亡数呈正相关 (P <0 0 5 )。结论 左旋四氢巴马汀可减少脑细胞凋亡 ,其机制与抑制缺血再灌注后脑组织钙聚集有关。

Abstract:Objective By combining observation variation calcium contents of brain tissues and cerebral cell apoptosis during cerebral ischemia reperfusion,we investigate the mechanism by which L-tetrahydropalmatine(L-THP) acts during cerebral ischemia reperfusion.Methods Use Pulsinelli's methods to create models of rats with cerebral is chemia reperfusion injury.Use atomic absorption spectrophotometer(AAS) to analysis calcium contents and use flow cytomyter(FCM) to analysis apoptosis.Results Calcium contents and the number of apoptotic cerebral cells in 12 h subgroup of cerebral ischemia reperfusion group was higher than sham-operation group(P<0.01),calcium contents and the number of apoptotic cerebral cells in 24 h subgroup of cerebral ischemia reperfusion group was more higher than sham-operation group(P<0.01). L-THP can control the rising of calcium contents of brain tissues and the number of apoptotic cerebral cells during both 12 h and 24 h of reperfusion(P<0.01).Variation of calcium contents in each group and variation in the number of apoptotic cerebral cells showed a marked positive relationship(P<0.05).Conclusion L-THP obviously reduced cerebral cell apoptosis caused by cerebral ischemia reperfusion injury,probably relating to inhibition of calcium aggregate.
