摘    要:目的 构建一个针对Graves病(GD)患者IgG的鼠单链抗体,体外实验证明其是否具有拮抗甲状腺刺激性抗体(TSAb)的能力.方法 利用噬菌体展示技术,得到一个抗TSAb的噬菌体抗体scFv,测定其相对亲和力.用表达重组人促甲状腺激素受体(hTSHR)的中国仓鼠卵巢细胞测定GD患者血中TSAb水平,并观察此scFv片段对GD患者IgGs诱导cAMP释放的抑制作用.结果 16例GD患者中有13例TSAb活性被scFv克隆T17显著抑制(最大抑制率为88%),相对亲和力为2.8 mol/L.结论 本实验利用噬菌体展示技术,筛选出一个具有较强拮抗TSAb活性的scFv克隆(T17).

Abstract:Objective To produce a mouse single chain Fv antibody fragment, and to evaluate whether it can act as a thyroid stimulating antibody (TSAb) antagonist. Methods We used phage-display technology to produce a scFv. Tested its relative affinity and effect on inhibition of cAMP production, which was induced by IgGs of patients with Graves' disease. And used Chinese hamster ovary cells transfected by human thyroid stimulating hormone receptor to detect the TSAb level of patients with Graves' disease.Results T17 was an antagonist of TSAb activity in 13 of 16 patients with Graves' disease, and its relative affinity was. 2.8 mol/L. Conclusion The scFv ( T17 ) produced by phage-display technology can act as a powerful TSAb antagonist.
