宁夏地产黄芩药材HPLC指纹图谱的研究

摘    要:目的建立宁夏黄芩药材HPLC-UV色谱指纹图谱,为鉴定黄芩药材、规范化种植提供实验数据。方法采用HPLC-UV方法对宁夏不同生长年限、产区的14个样品黄芩药材建立指纹图谱。色谱柱Thermo ODS-2HYPERSIL(250mm×4.6mm,5μm);甲醇-乙腈-醋酸-醋酸钠(pH=4.5)缓冲液为流动相进行梯度洗脱;检测波长276nm,流速1ml/min,柱温25℃。结果建立了宁夏黄芩药材HPLC-UV指纹图谱,标定出18个共有峰,计算出各批次药材间的相似度。结论各批黄芩药材指纹图谱相似度大小与其裁培地区、生长年限、采收期等因素相关,所建立的指纹图谱可作为黄芩药材的鉴定方法和宁夏黄芩药材规范化种植的实验依据。

Abstract:Objective To establish chromatographic fingerprint of Scutellaria baicalensis by HPLC which could be used to Scutellaria baicalensis's identity and standard planting.Methods A method of HPLC-UV was applied to establish chromatographic fingerprint of 15 Scutellaria baicalensis samples from different region and growth in Ningxia.The chromatographic condition Thermo ODS-2HYPERSIL(250mm×4.6mm,5μm) column,the mobile phase consisted of methanol,acetonitrile,and sodium acetate-acetic acid buffer(PH=4.5) which was in the gradient elution at a flow rate of 1.0 mL·min-1,at 25℃.Results Established the chromatographic fingerprint of Scutellaria baicalensis in Ningxia,18 peaks were identified as the characteristic fingerprints of Scutellaria baicalensis.The similarity of Radix Astragali from different region and growth was caculated.Conclusion The similarity of Scutellaria baicalensis is related with its' region and growth.This fingerprint was useful for the identity and standard planting of Scutellaria baicalensis.
