摘    要:目的 探讨胚胎脊髓移植并神经营养因子防止成年大鼠脊髓轴突损伤后引起神经元萎缩的作用。方法 采用大鼠腰脊髓半切洞损伤模型,将实验动物分为6组:A组:单纯脊髓损伤组,B组;胚胎脊髓移植组;C组:损伤+移植+NGF组;D组:损伤+移植+CNTF组;E组:损伤+移植+NMT-3组;F组:损伤+移植+BDNMF组。手术后应用行为学和电生理检查观察大鼠后肢功能恢复情况,应用Nissl染色方法观察脊髓神经元的大

Effects of transplantation of fetal spinal cord and neurotrophic factors on neurons of injured spinal cord in adult rats

Abstract:Objective To study the effects of the transplantation of fetal spinal cord and neutrotrophic factors on the reversion of axonotomy induced atrophy of the neurons in the injured spinal cord of adult rats. Methods Hemisection of the lumbar segment of the spinal cord was performed for all the experimental rats. The effects of neurotrophic factors NGF, CNTF, NT 3 and BDNF were studied. The rats were divided into 6 groups as follows: Group A with hemisection of the cord only; Group B with cord hemisection and fetal cord grafting; Group C with cord hemisection, fetal cord grafting and NGF; Group D with cord hemisection, fetal cord grafting and CNTF; Group E with cord hemisection, fetal cord grafting and NT 3; and Group F with cord hemisection, fetal cord grafting and BDNF. Combined behavioral scores (CBS), somatosensory evoked potentials (SEP) and motor evoked potentials (MEP) were measured. The slides with Nissl staining were used to measure the size of the neurons. The positive cells were quantitatively analyzed with a computer image analysis system. Results The size of the neurons of the 6 groups was arranged in the order from large to small as F>E>D>C>B>A ( P <0.05). The recovery of the injured neurons was best in Group F as evidenced by morphological observation. The increase of the size of neurons was correlated to their functional recovery. Conclusion The combination of the transplsntation of fetal spinal cord and neurotrophic factors can promote the structural and functional recovery of the injured neurons.
