β防御素在肺气虚证大鼠发病中的作用

摘    要:目的观察肺气虚证大鼠血清β防御素和支气管灌洗液、肺匀浆β2防御素水平的变化,研究β防御素在肺气虚证大鼠发病中的作用。方法采用呼吸道生物被膜菌(铜绿假单胞菌)滴鼻法引起大鼠呼吸道慢性感染,辅以寒冷、疲劳刺激,诱导肺气虚证的发生。分别在大鼠感染后3,30,100d观察肺组织病理学变化,酶联免疫吸附试验测定其血清β防御素,气管冲洗液、肺匀浆&防御素水平。结果模型组大鼠在感染后3d,肺部急性感染明显,血清β防御素、肺匀浆β2防御素水平显著升高;感染30,100d时大鼠肺部显示慢性感染,血清β防御素和气管灌洗液、肺匀浆β2防御素水平显著降低(P〈0.01)。结论β防御素参与了肺气虚证的发病过程,发病急性期参与炎症反应,慢性期诱发或加重肺气虚证。

Abstract:Objective To observe the changes of serum contents of β-defensin,and the levels of β2-defensin in bronchial lavage fluid and lung homogenates,and investigate the effects of β-defensin on lung-qi deficiency syndrome in rats.Methods Nasal drip of biofilm bacteria(Pseudomonas aeruginosa) combined with cold stress and exhaustive swimming was employed to develop lung-qi deficiency syndrome.Histopathological changes of lung were observed 3,30,100 days after infection,respectively.Enzyme linked immunosorbent assay was used to assay the serum contents of β-defensin,and the levels of β2-defensin in bronchial lavage fluid and lung homogenates.Results An obviously acute pulmonary infection was observed in model rats 3 days after infection,the serum contents of β-defensin and the levels of β2-defensin in lung homogenates were significantly increased.Chronic pulmonary infection was developed 30,100 days after infection,respectively;the serum contents of β-defensin and the levels of β2-defensin were significantly decreased.Conclusion β-defensins were involved in pathogenesis of lung-qi deficiency syndrome;it participates in inflammatory reaction during acute phase,and induces and aggravates lung-qi deficiency syndrome during chronic phase.
