肾衰2号方抑制残余肾TGF-β1与CTGF mRNA的过度表达

目的：探讨肾衰2号方通过抑制转化生长因子-β1（transforming growth factor beta1,TGF-β1）、结缔组织生长因子（connective tissue growth factor,CTGF）mRNA的过度表达而减轻肾脏纤维化的作用机制。方法：制作大鼠5/6肾切除慢性肾衰竭大鼠肾纤维化模型,分别给予肾衰2号方及主要单味药党参、桃仁灌胃,并设科素亚对照组。治疗2个月后用RT-PCR的方法检测CTGF和TGF-β1 mRNA的表达。结果：在5/6肾切除慢性肾衰竭肾纤维化模型组中表现为TGF-β1、CTGF mRNA的过度表达（P〈0.01）,经肾衰2号方治疗后TGF-β1、CTGF mRNA的表达降低（P〈0.01）,科素亚也有一定的作用,而党参、桃仁的作用不明显。结论：肾衰2号方减轻肾纤维化的机制在于抑制细胞因子TGF-β1、CTGF mRNA的过度表达。

Inhibition on Excessive Expression of TGF-β1、CTGF mRNA by No.2 Renal Failure Recipe in Remnant Kidney

Objective：To investigate effect of No. 2 Renal Failure Recipe on reducing renal fibrosis by inhibiting the overexpression of the TGF-β1 and CTGF mRNA. Methods：The models of 5/6 nephrectomized rats were prepared and treated with No. 2 Renal Failure Recipe, radix codonopsitis and peach seed respectively. After two months, connective tissue growth factor and transforming growth factor- beta 1 mRNA were examined by RT- PCR. Results：Over- expression of TGF-β1 and CTGF mRNA were observed in the model group （P〈0.01） ; but after 2 month treatment of No. 2 Renal Failure Recipe,the expressions declined significandy （P〈0.01）. Cozaar had similar effect at some degree, but radix codonopsitis or peach seed did not. Conclusion：The mechanism of No.2 Renal Failure Recipe on reducing renal fibrosis is attributed to inhibit cytokine over- expression of TGF - β1 ,CTGF mRNA.
