胸水细胞8-OH-dG,k-ras基因突变在肺癌诊断中的意义

摘    要:目的探讨胸水细胞8-OH-dG,k-ras基因蛋白表达和突变与肺癌的关系.方法采集53例肺癌病人、53例非肺癌病人的胸水,离心得细胞,将细胞成分离心涂片,用免疫组化学分别测定8-OH-dG,k-ras基因的蛋白表达和用PCR-SSCP法检测胸水细胞k-ras基因突变率.结果肺癌病人胸水细胞8-OH-dG,k-ras基因的蛋白表达阳性率分别为77.4%(41/53)与56.6%(30/53);非肺癌病人胸水细胞8-OH-dG,k-ras基因的蛋白表达阳性率分别为13.2%(7/53)与3.8%(2/53),两组比较差异有高度显著性(P<0.01);PCR-SSCP法检测肺癌病人、非肺癌病人胸水细胞,k-ras基因突变为62.3%(33/53)与5.7%(3/53),两组比较差异有高度显著性(P<0.01).53例肺癌病人胸水细胞8-OH-dG,k-ras基因的蛋白表达分值分别为1.68±1.21与1.37±1.13,等级相关分析呈高度正相关(Rs=0.743,P<0.01).结论肺癌病人胸水细胞,8-OH-dG,k-ras基因的蛋白表达阳性率、分值以及PCR-SSCP法检测胸水细胞k-ras基因突变率均高于非肺癌病人;肺癌组胸水细胞8-OH-dG,k-ras基因蛋白表达两者有关联.

Abstract:Objective:To investigate the relationship between the expression of8-OH-dG and k-ras gene protein in pleural cell of lung cancer group and control group.Methods:Pleural of53cases of lung cancer and53cases without lung cancer were selected.The pleural cell was separated from the pleural by cen-trifuge.The cells were spread on a slide by centrifuge.Immunohistochemical PCR-SSCP was used to detect rate of k-ras gene mutation in lung cancer group and control group.Results:8-OH-dG and k-ras gene pro-tein were both expressed in lung cancer group and control group,the positive rate of8-OH-dG were77.4%(41/53)and13.2%(7/53),the positive rate of k-ras were56.6%(30/53)and3.8%(2/53).Compared the lung cancer group with control group in8-OH-dG and k-ras,differences were both statistically significant(P<0.01).Rate of PCR-SSCP deteced k-ras gene mutation in lung cancer group were62.3%(33/53)and controlgroup were5.7%(3/53),differences were both statistically significant(P<0.01).A high positive correlation existed between the expressions of8-OH-dG and k-ras protein in lung cancer by degree relative analysis(Rs=0.743,P<0.01).Conclusion:The positive rates of8-OH-dG and k-ras proteins expressed in pleural cell of lung cancer group were higher than those in pleural cell of control group.The expressions of8-OH-dG and k-ras proteins are both closely related in pleural cell of patients with lung cancer.
