摘    要:目的采用RAPD分子标记技术对不同产地美花石斛进行分析,为美花石斛种质资源的准确鉴别提供依据。方法用RAPD分子标记技术对不同产地美花石斛基因组总DNA进行PCR扩增,根据其扩增产物分子量大小进行编码计算。结果从117种随机引物筛选出8种有效引物,S2108的PCR扩增产物中的特异性位点能够鉴别出广西玉林居群和贵州安居群。结论利用RAPD分子标记技术鉴别美花石斛种质资源是可行的,在严格控制各项条件下能够保证鉴别美花石斛种质资源的稳定性和重复性。

Abstract:Objective To authenticate the germplasm resources of Dendrobium loddigesii collected from different areas by RAPD.Methods The total DNA of D.loddigesii was amplified by RAPD-PCR,and the amplified DNA were investigated based on their molecular weight and amplifying distances.Results 8 effective primers were selected from 117 random primers.Some polymormic loci amplified from primer S2108 could effectively identify Yulin population and Anlong population of D.loddigesii.Conclusion It is feasible for identifying germplasm resources of Dendrobium loddigesii using RAPD molecular marker,and the stability and repeatability of identification results can be insured under the strict and normative experiment processes in RAPD.
