脑络欣通影响气虚血瘀证脑缺血再灌注模型大鼠海马CA1区神经元凋亡的机制研究

摘    要:目的:探讨中药复方脑络欣通改善脑缺血损伤的作用机制。方法:采用多因素复合制作气虚血瘀证大鼠脑缺血动物模型,观察中药脑络欣通对脑缺血-再灌注损伤的预防与治疗作用。用免疫组化方法观察缺血-再灌注模型大鼠海马CAI区Bel-2蛋白、Bax蛋白及脑源性神经营养因子的表达;用TUNEL法观察缺血.再灌注模型大鼠海马CAI区神经元的凋亡现象;用硫代巴比妥酸比色法测定海马匀浆丙二醛浓度。结果:(1)预防及治疗给药可以减少缺血-再灌注模型大鼠海马CAI区神经元的凋亡现象,并可以使缺血-再灌注模型大鼠海马CA1区Bcl-2蛋白及脑源性神经营养因子表达升高,使Bax蛋白的表达减少;(2)预防组可以降低缺血急性期病理性升高的丙二醛含量。结论:脑络欣通可以减少缺血-再灌注模型大鼠海马CA1区神经元的凋亡现象,其机制可能与减少脑缺血-再灌注后自由基损伤有关,并通过影响凋亡相关蛋白的表达减少神经元凋亡。

Abstract:Objective: To study the mechanism of Naoluoxintong in preventing and treating on cerebral ischemia and reperfusion damage.Methods: The cerebral ischemia and reperfusion model with Qi-deficiency and blood stasis syndrome was established to investigate the preventive and therapeutical effect. The expressions of Bcl-2 protein and Bax protein were observed by the method of immunohistochemistry; the apoptosis was observed by the method of TUNEL Results:Naoluoxintong can counteract neural cell apoptosis after cerebral ischemia and reperfusion;Naoluoxintong can increase the expression of Bcl-2 protein while reduce the expression of Bax protein. Conclusions: Naoluoxintong can counteract neural cell apoptosis after cerebral ischemia and reperfusion with the effection on apoptosis-related protein.
