巴泰病毒云南株生物学性状研究

摘    要:摘要: 目的 掌握巴泰(Batai)病毒生物学特性,为巴泰病毒的监测和调查研究提供科学依据。方法 用分离自云南省菲律宾按蚊(Anopheles philippinensis)的巴泰病毒LC92-4株,采用细胞培养、小白鼠感染、血凝抑制、间接免疫荧光、空斑试验以及病理检测和电镜观察等方法进行生物学特性试验。结果 巴泰病毒可引起<4周龄小白鼠发病死亡。感染鼠脑病毒颅内、皮下和腹腔接种2周龄小白鼠的LD50分别为5.67logLD50/0.025mL、5.3logLD50/0.03mL和4.5logLD50/0.03mL。感染病毒鼠的心、肝、脾、肺、肾病毒悬液颅内接种2周龄小白鼠,LD50分别为3.75、4.25、2.25、3.0、2.25 logLD50/0.025mL。感染发病小白鼠脑、心、肝、脾、肺、肾均发现病理改变。感染鼠的脑组织经电镜检测发现圆形有包膜病毒颗粒,大小约82nm。巴泰病毒能在BHK21和Vero细胞产生明显病变,并可产生空斑;在BHK21细胞上,空斑滴度为6.04logPFU/mL;在C6/36细胞上不产生病变。感染病毒鼠脑接种BHK21和Vero细胞,TCID50分别为 5.50logTCID50/0.025mL和5.00 logTCID50/0.025mL。在pH5.75~7.0条件下,巴泰病毒不具有与鸽、鹅、鸡、鸭、人O型和绵羊红血球发生凝集的特性;空斑减少中和试验检测云南省357份人血清,阳性2份,中和抗体滴度均为1:10。结论 乳小白鼠和幼年小白鼠对巴泰病毒高度易感。BHK21和Vero细胞对巴泰病毒敏感,可用于病毒分离培养和空斑测定。建立的免疫荧光和空斑减少中和试验具有特异性,可用于巴泰病毒抗体检测。应进一步开展该病毒的分布及其与疾病关系的研究。

Study on biological characters of Batai virus isolated from Yunnan province
Abstract:ABSTRACT: The purpose of this study is to discover the biological characteristics of Batai virus isolated from Anopheles philippinensis in Langcan county, Yunnan province and nominated as LC94-2, including those of susceptibility of mice and tissue-culture cells and serological reaction. The mice younger than 4 weeks were susceptible to Batai virus. The titers of LD50 in 2-weeks old mice inoculated via endermic, intraperitoneal and intracerebral routes were 4.5logLD50/0.03ml, 5.3logLD50/0.03ml and 5.67logLD50/0.025ml, respectively. The tissue suspensions of heart, liver, spleen, lung and kidney of morbid mice were inoculated with 2-weeks old mice and the titers of LD50 were 3.75、4.25、2.25、3.0、2.25 logLD50/0.025mL. Pathological changes were found in the organs. Virus particles were found in the infected mice brain via electron microscope detection and the size of them were about 82nm. CPE was observed in BHK21 and Vero cells, and not in C6/36 cell. The titers of TCID50 in BHK and Vero were respectively 5.50logTCID50/0.025ml and 5.00 logTCID50/0.025ml. PFU in BHK21 was 6.04logPFU/ml. No hemagglutinative character was observed with the six kinds of red blood cells. As buffer solutions were chosen ranging from pH5.75-pH7.0, no hemagglutinative character was observed with the six kinds of red blood cells. With plaque reduction neutralization test, 2 positive sera among 357 human sera from Yunnan province were detected with the same titer of 1:10. Our results show that the suckling and infant mice are highly susceptible to BATV. BHK21 and Vero cells, both of which are susceptible to BATV, can be employed for isolating this virus and plaque assay. Both of IFA and PRNT are applicable to detect the antibody of this virus.
