凝结芽孢杆菌TBC-169菌株治疗抗原诱导大鼠结肠炎的实验研究

摘    要:背景:开发新的溃疡性结肠炎(UC)治疗药物是当前研究的热点。目的:探讨凝结芽孢杆菌(B.coagulans)TBC-169菌株对抗原诱导的大鼠结肠炎的疗效和作用机制。方法:以牛结肠黏膜蛋白多次免疫建立大鼠结肠炎模型,予B.coagulansTBC-169菌株制剂和美沙拉秦单独或联合治疗。21d后处死大鼠,评定结肠湿重指数、溃疡指数和组织学评分,测定肠系膜淋巴细胞转化率和血清白细胞介素(IL)-8、肿瘤坏死因子(TNF)-α、IgG水平。结果:美沙拉秦组、大剂量B.coagulans组和联合治疗组结肠湿重指数较模型对照组显著降低(P〈0.05)。各治疗组结肠溃疡指数和结肠中上段组织学评分较模型对照组显著降低,T细胞转化率显著升高,血清IL-8水平显著降低(P〈0.05)。与模型对照组相比,大剂量B.coagulans组和联合治疗组血清TNF-α水平显著降低(P〈0.01),小剂量B.coagulans组血清IgG水平显著降低(P〈0.01),单用美沙拉秦则无类似作用。结论:B.coagulansTBC-169菌株可明显改善模型大鼠的结肠黏膜损伤,其机制与调控炎症因子表达、恢复机体免疫功能等有关。

Abstract:Background: Development of novel drug is a hotspot in the study of ulcerative colitis (UC) in recent years. Aims: To investigate the therapeutic effect and mechanism of Bacillus coagulans (B. coagulans) TBC-169 strain in the treatment of antigen-induced colitis in rats. Methods: Colitis model was established by muhiple immunization of calf colonic mueosat protein (CCMP) in rats. The model rats were administrated with a preparation of B. coagulans TBC-169 strain and mesalazine in combination or separately for 21 days. Then all animals were sacrificed, the colonic wet weight index, ulcerative index and histologie score were evaluated, and the transformation rate of mesenteric lymphocytes and serum levels of interleukin (IL)-8, tumor necrosis factor (TNF)-α and IgG were determined. Results: The colonic wet weight index in mesalazine group, 10^8 CFU/ml B. coagulans group and combination group decreased significantly as compared with that in the colitis model controls (P〈0.05). When compared with the colitis model control group, the ulcerative index and histologic score of colonic mucosa in upper and middle segment of colon in all treated groups decreased significantly, whereas the transformation rate of T-lymphoeytes increased and serum level of IL-8 decreased significantly (P〈0.05). In 10^8 CFU/nd B. coagulans group and combination group, the serum level of TNF-α decreased significantly (P〈0.01), and in 107 CFU/ml B. coagulans group the serum level of IgG decreased significantly (P〈0.01) in comparison with those in the colitis model controls. No effect was observed on the serum levels of TNF-α and IgG in mesalazine group. Conclusions: B. coagulans TBC-169 strain can improve the colonic mucosal damage by modulating the expression of inflammatory cytokines and restoring the immunologic function in colitis model rats.
