摘    要:转基因动物的鉴定是转基因动物研究领域的重要内容。本文简要回顾了相关鉴定技术的研究进展 ,建立和完善了特异性PCR扩增及其产物酶切的转基因动物鉴定体系 ,在引物设计、PCR参数优化、设立内源性标记等方面作了进一步的改进 ,提高了PCR的特异性和可靠性。实验表明 ,PCR扩增及其产物酶切是一种准确、灵敏、简便、重现性好的转基因动物筛选鉴定方法

Abstract:Identification of transgenic animals plays an essential role in transgenic study. In this report, the technical progress for screening transgenic offspring was briefly reviewed and a simple and efficient assay was introduced. Transgenic animals were identified and confirmed by PCR along with further restriction endonuclease analysis. Some factors on PCR, such as the design of primers, the concentration of Mg+{2+} and DMSO, the annealing temperature and so on, were optimized. In order to assess the validity of the template, a pair of primer which is specific for the endogenous beta-glolin gene was used as control. The results showed that the assay was a reliable, sensitive, rapid and well-repeated to identify transgenic animals.
