摘    要:目的]探讨IL-37在重症急性胰腺炎小鼠模型中的动态变化及意义。方法]采用牛磺胆酸钠注射诱导小鼠重症急性胰腺炎,再用乌司他丁(UTI)给予治疗,分别于6h、12h、24h时处死小鼠,留取血清和胰腺组织。ELISA法检测各组小鼠血清中TNF-α、IL-6、TGF-β1和IL-37表达水平,利用Western blot和Real-Time PCR方法检测小鼠胰腺组织IL-37蛋白及IL-37 mRNA表达水平。结果]重症急性胰腺炎组(SAP组)小鼠血清细胞因子TNF-α和IL-6水平均随着时间延长而显著增加(P〈0.01);TGF-β1和IL-37的表达水平在12h后明显降低(P〈0.01)。胰腺组织中IL-37蛋白及IL-37 mRNA表达也在12h后显著降低(P〈0.01)。而经UTI治疗后,小鼠血清TNF-α和IL-6水平均随时间延长而降低,而TGF-β1和IL-37的表达水平则随时间延长而增高,并且胰腺组织中IL-37蛋白和IL-37 mRNA的表达水平也相应增高(P〈0.01)。结论]SAP小鼠中IL-37可能作为抑炎性细胞因子参与机体的病理变化。

Study on the Dynamic Changes of IL-37 in Severe Acute Pancreatitis in Mice and Its Significance

Abstract:Objective] To investigate the dynamic changes and the significance in the mice model with severe acute pancreatitis.Methods] Induced by sodium taurocholate into mice with severe acute pancreatitis as SAP and ulinastatin treatment with SAP as the UTI group of mice.Saving serum and pancreatic tissue samples after the mice were sacrificed at 6h,12h,24h respectively.TNF- α,IL-6,TGF-β1 and IL-37 in serum were tested by ELISA.And Western blot and Real-Time PCR were used to test IL-37 protein content and IL-37 mRNA levels in pancreatic tissues.Results] SAP serum TNF- α and IL-6 levels in SAP mice significantly increased with time.Serum TGF-β1 and IL-37 levels decreased obviously after 12h(P〈0.01).IL-37 protein contents and IL-37 mRNA levels were significantly decreased after 12h(P〈0.01).After treated with ulinastatin,TNF-α and IL-6 levels decreased while TGF-β1 and IL-37 levels increased significantly and IL-37 protein and mRNA expressions in pancreatic tissues also increased(P〈0.01).Conclusion] In SAP mice,IL-37 might play a role as an anti-inflammatory cytokine in pathophysiology mechanisms in SAP mice.
