川芎嗪对二甲酰胺引起的PC12细胞损伤的保护作用

摘    要:目的分析川芎嗪对二甲酰胺引起大鼠肾上腺嗜铬细胞瘤克隆化细胞株PC12细胞损伤的保护作用,探讨川芎嗪治疗脑缺血再灌注损伤的机制。方法采用川芎嗪预处理PC12细胞后,制备二甲酰胺细胞损伤模型,CCK-8法检测细胞毒性、流式细胞术分析Caspase-3、8、9、Western blot检测HMGB1、ELISA检测氧化应激指标观察二甲酰胺的毒性及川芎嗪的保护作用。结果川芎嗪预处理后,PC12细胞的存活数显著提高,Caspase-3和Caspase-9活性降低,未见HMGB1表达降低,SOD上调,LDH和MDA下调,GSH升高。结论川芎嗪对二甲酰胺引起的PC12细胞损伤有显著的保护作用,但其保护作用可能与川芎嗪抑制细胞凋亡、调节氧化应激反应相关。

Abstract:Objective To analyze whether tetramethylpyrazine could protect PC12 cell injury induced by diamide in order to explore the mechanism of tetramethyipyrazine in the treatment of cerebral ischemia-reperfusion injury.Methods After PC12 cells were cultured by tetramethylpyrazine,diamide was added to induce the injury of PC12 cells.The toxicity of diamide and the protective effects of tetramethylpyrazine were detected and evaluated.Cytotoxicity was detected by CCK-8.Activity of caspase-3,8,9 was detected by flow cytometry.HMGBl was detected by Western blot.Oxidative stress was detected by ELISA.Results After the pre-treatment with tetramethylpyrazine,the survival rate of PC12 cell was significantly improved.The activity of Caspase-3 and Caspase-9 was reduced.The expression of HMGBl was not decreased.Meanwhile,the expression of SOD and GSH were increased,and the LDH and MDA decreased.Conclusion Tetramethylpyrazine has a significant protective effect on PC12 cells injury caused by diamide.Its protective effect may be related to inhibit apoptosis and regulate oxidative stress.
