体外震波对家兔肝组织影响的硝酸镧示踪和电镜酶细胞化学研究

摘    要:采用体外震波碎石机对家兔肝脏实施震波冲击实验,并用硝酸镧示踪和碱性磷酸酶细胞化学亚显微定位技术对兔肝组织进行电镜研究。观察显示:实验组肝见镧颗粒穿过细胞膜进入胞质,并围绕线粒体呈颗粒状沉积或选择性地进入线粒体;毛细胆管腔及紧密联接区和肝细胞间隙均有镧颗粒沉积,紧密联接区及肝细胞间隙亦见异常的碱性磷酸酶反应物。超微结构示线粒体肿胀、膜破坏及内质网扩张。结果表明,震波作用的靶细胞单位是膜结构和线粒体,其早期损伤效应是细胞膜性结构的通透性增高及完整性的丧失,进而导致亚细胞结构改变和生化的异常。

Abstract:Tissue effects in the liver of rabbit undergoing extracorpeal shock wave lithotripsy(ESWL) were studied with Electron Microscope.THe application of lanthanum as a marker for ultrastruc- tural study demonstrated that intracellular lanthanum could be observed and it entered the swollen mitochonndria selectively.The ALPase reaction products and lanthanum granules were both found to deposite in the zone of tight junctions of bile apillaries. Ultrastructurally,The or- ganelle changes of the liver commonly seen included hydropic mitochondria,frank cavitation of the mitochondria matrix and loss of cristae,dilatation and vesiculation of endoplasmic reticulum. The results showed that the early effects of ESWL on the liver were an increase in permeability and a loss of integrity of the membranous organelles of the liver cells.
