应用基因芯片探讨天芪降糖胶囊降糖调脂的机制

摘    要:目的探讨天芪降糖胶囊改善血糖和血脂的机制。方法选用KKAy小鼠16只,随机分为天芪组(给予天苠降糖胶囊)和对照组(给予生理盐水),灌胃4周。测定FBG、血脂和Fins,并行OG—TT。取小鼠骨骼肌进行RT-PCR基因芯片实验。结果天芪组FBG、OGTT曲线下面积、HOMA-IR、TC和TG均比对照组显著降低(P〈0.05或P%0.01)。基因芯片显示天芪组GluT-4、MAPK-8、MAPK-14和PPARq上调,PPAR-γ、PGC、CEBP、TNF—α、NF-κB、ICAM-1和IL-6下调。结论天芪降糖胶囊能有效降低KKAy小鼠FBG,调节脂代谢。血糖降低机制可能与MAPK通路和GluT-4上调有关;另外可能通过下调炎症因子水平改善胰岛素抵抗。血脂改善可能与PPAR—α上调有关。

Abstract:Objective To explore the effects of Tianqi capsule on the FBG, lipid metabolism and fasting insulin(Fins) level in KKAy mice and to investigate their possible therapeutic mechanisms. Methods KKAy mice were randomly divided into two groups: Tianqi group(treated with 1.5g/kg · d Tianqi) and control group(treated with the same volume of NS). for 4 weeks. FBG, TC, TG, HDL-C, LDL-C and Fins were detected. The OGTT test was done. RT-PCR array test was done with skeletal muscle. Results Tianqi capsule could decrease FBG, glucose area under the curve (AUC), HOMA-IR, TC and TG, compared with those of control group(P〈0. 05 or P〈0. 01). RT-PCR array test results indicated that Tianqi up-regulated the expression of GLUT4, MAPK8, MAPK14 and PPARa, while the expressions of PPART, PGC, CEBP, TNF~, NFgB, ICAM-1 and IL-6 were down-regulated. Conclusions Tianqi capsule possesses the functions of regulating the levels of blood glucose and lipids in KKAy mice. And the glucose-lowing mechanisms are correlated with GLUT4 and MAPK pathway. And up-regulated PPARa involves lipid improvement. Moreover, Tianqi capsule can decrease inflammatory cytokines to improve insulin resistance.
