库拉索芦荟多糖的分离纯化和含量测定

摘    要:目的分离纯化库拉索芦荟多糖并建立含量测定方法.方法醇沉法提取库拉索芦荟多糖,Sephadex G-100凝胶柱色谱纯化.以甘露糖为标准,苯酚-硫酸法测定库拉索芦荟多糖含量.结果经分离纯化得到的主要组分库拉索芦荟多糖Ⅰ含量为81.11%,测定平均回收率为100.3%.结论该方法可从库拉索芦荟中提取分离纯度较高的多糖,含量测定方法简便、快速、重现性好.

Abstract:OBJECTIVE To isolate, purify the polysac ch aride of Aloe barbadensis Miller and establish the content determination method.METHOD The polysaccharide was precipitated with ethanol and fur ther purified on Sephadex G-100 column. Its total soluble sugar content was det ected colorimetrically by the phenol-sulfuric acid method with mannose as stand ard. RESULTS The total sugar content of polysaccharide I was 81.11%. The average recovery was 100.3%. CONCLUSION It is possible to extract and isolate purified polys accharide from Aloe barbadensis Miller. And the determination method is simple, rapid and well reproducible.
