摘 要: | 目的:研究大鼠中度(0.2MPa)侧位液压冲击脑损伤时大脑皮质立即早期基因c-fos和c-jun表达产物Fos蛋白和Jun蛋白的变化规律。方法:雄性SD大鼠,随机分为正常对照物、手术对照组和损伤组。损伤组动物均给以0.2MPa液压冲击脑损伤,按冲击后处死时间不同再分为5、15、30、60、120、240、480和720分钟组。应用免疫组织化学方法观察Fos和Jun蛋白在大脑皮质的表达特点。结果:冲击后30分钟,双侧大脑皮质Fos阳性细胞数逐渐增多,冲击后720分钟达高峰。Fos阳性细胞面积在冲击后120分钟逐渐增大,720分钟达高峰;冲击后60分钟双侧大脑皮质Jun阳性细胞数逐渐增多。冲击后120分钟阳性细胞面积逐渐增大,冲击后720分钟阳性细胞数和面积均达高峰。结论:中度侧位液压冲击脑损伤后Fos蛋白和Jun

Expression of c-fos and c-jun in cerebral cortex following moderate lateral hydraulic percussion brain injury in rats |

Abstract: | Objective Toinvestigate the protein expression of cfos and cjun in cerebral cortex following moderatelateral fluid percussion brain injury(0. 2 MPa) in rats.MethodsMale SpragueDawley ratswere divided into normal control,sham o peration control and injury group.Animals ininjury group were subjected to a m oderate lateral hydraulic percussion brain injury (0.2MPa),and then were subdi vided into 5,15,30,60,120,240,480 and 720 minutes groupsaccording to the tim e elapsed after injury.The expression of cfos and cjun w asdetermined by immunohistochemistry. ResultsThirty minutes after percussion,Fos positiveneurons increase d bilaterally and progressively in cerebral cortex,peaking at 720minutes.The a rea of Fos positive neurons elevated at 120 minutes after percussion,peakingat 720 minutes.Jun positive neurons increased bilaterally at 60 minutes after percussion,and remained elevated up to 720 minutes after brain injury.The area of Jun positiveneurons increased at 120 minutes af ter percussion,peaking at 720 minutes. ConclusionsTheinduction and expression of cf os and cjun in cerebral cortex increased bilaterally aftermoderate hydraulic percussio n brain injury,and lasted for a long period.The elevatedinduction of Jun appea rs to be delayed as compared with Fos,but the temporal pattern andexpression e xtent might be similar to that of Fos. |
